yeah, read the release notes, there's a link on the download page to them.
you need to install the cuda 3.0 drivers and cuda toolkit.
on 10.5 you also need to download and install XQuartz, as stated on the downloads page.

any reason why you're not running the last Leopard update (I think it was 10.5.8) on your Macs?
as for CUDA install, I don't remember having such a problem, I think I made a plain standard installation in my system HD... but I'm on Snow Leopard.

as far as I can tell any free 10.5.x update should be safe (think about them as automatic update or Service Packs under Windows, while upgrading from 10.5.x to 10.6.x is like going from Vista to 7, for example). they are usually about bug fixes and better hardware/software driver and support (and much more, I know...)
you have not said what Graphic Cards you have in your Macs, that should help to see what's wrong.
another thing that come to mind is Uninstall the not-running previous Octane installation before installing CUDA drivers, then Toolkit and finally Octane demo.
EDIT: one more thing. after CUDA install you should see a CUDA preference button in "Preferenze di Sistema - Accessori" (see screnshot)

this is very strange indeed! is it happening with CUDA install only? any chance you have run out of space or you accidentally marked your HD/partition as read only?

I don't know since I switched to Snow Leopard before doing any CUDA installation, but what you said makes sense. if so you can try to ask at the nvidia forum: http://forums.nvidia.com/ - there's a "CUDA on OSX" section - or upgrade to 10.5.8 (maybe make a time machine backup before, if you're not sure)
